STARR-seq assay performed on uncharacterized cis-regulatory elements to find candidate silencer elements in K562 cells. Overall design: Five wells of 6 well plates, each with about 1 million K562 cells, were transfected with 20 ug of plasmid containing our library. Cells were grown in antibiotic-free media for 24 hours and then harvested for RNA and DNA extraction.
