This study aimed to investigate the effect of glucose restriction (GR) on energy metabolism and muscle fiber type in skeletal muscle. To achieve this goal, we constructed a mouse model of innate glucose restriction by mutating the glucose transporter 4 (Glut4), the major glucose transporter in skeletal muscle. A glutamine-to-leucine mutant was introduced by CRISPR/Cas9-mediated gene-editing in the QL motif in the 5th transmembrane domain of mice Glut4 (Glut4 Q177L), resulting in blocked substrate binding and the consequent glucose restriction in skeletal muscles. Transcriptome sequencing was performed on gastrocnemius samples from homozygous Glut4 mutant mice (Glut4m) and wild-type C57BL/6 mice (WT) at four growth stages, including young (12 weeks), adult (24 weeks), old (36 weeks) and very old (60 weeks).
