FGF23 is a biomarker associated with morbidity and mortality, but controversies remain if a causative link exists between FGF23 excess and adverse renal consequences in chronic kidney disease (CKD). Here, we investigated renal FGF23 signaling in kidney disease models in mice, with a focus on adverse pro-inflammatory effects assessed in bulk transcriptomes. To investigate renal effects of FGF23 hormone in kidney disease, we assessed bulk transcriptomes in kidney of FGF23 or vehicle treated mice that were i) healthy males or males with anti-glomerular basement membrane disease receiving 6 days of treatments, ii) healthy females or females with adriamycin nephroathy receiving 24h of treatment, and iii) adenine nephropathy mouse kidney tissue receiving ex vivo treatment for 1h in precision-cut kidney slices (PCKS).
