Real-time quantitative PCR profiling of human miRNAs in CAG RNA-expressing cells

We examined whether PAPD5 upregulation induced by mutant CAG RNA led to any dysregulation of cellular miRNA profiling. We found that, out of the 751 miRNAs examined, the expression of 186 miRNAs was reduced in EGFPCAG78-expressing human neuroblastoma SK-N-MC cells and was restored back to normal levels upon the knockdown of PAPD5 expression. qPCR gene expression profiling. Equal amount of small RNAs from each sample was used for gene expression analysis. The experiments were repeated independently for three times.
