High throughput sequencing of gene editing efficiency in P. acidilactici

In order to establish the genome editing tool, we selected two target sites respectively on pyrE gene. Transformants on the plates were randomly selected and incubated in the liquid medium. The target regions were PCR amplified, and the agarose gel analysis indicated that genome editing has happened. After one-round passage in the fresh medium, the amplification products were subjected to high throughput sequencing, the quantification of in vivo gene editing efficiency was obtained.
