In order to identify RNA binding proteins that are essential for male gametogenesis in the rodent malaria parasite P. yoelii, we conducted RNA-seq analysis to identify RBPs that are specifically expressed in male gametocytes. Overall design: A double fluorescence reporter strain DFsc7, which allowed for specific expression of GFP in male gametocytes and mCherry in female gametocytes, was utilized to induce gametocytes in mice. Subsequently, the male and female gametocytes were sorted using fluorescence-activated cell sorting (FACS), and RNA-seq analysis was conducted to generate gender-specific transcriptomes. Addition: Through Principal Component Analysis (PCA) analysis, the initial sample, Male biol rep1, showed poor reproducibility in RNA-seq data. Subsequently, we re-sorted and obtained three samples of male gametes (Py17XNL_MG_Rep1, Py17XNL_MG_Rep2, and Py17XNL_MG_Rep3) for RNA-seq, which yielded more reliable data.
