This project aims to generate a high-quality, telomere-to-telomere (T2T) genome assembly of Cannabis sativa using a combination of long-read sequencing, short-read Illumina sequencing, full-length transcriptome data, and Hi-C scaffolding technologies. High-molecular-weight genomic DNA was sequenced using Oxford Nanopore and PacBio HiFi platforms to achieve both ultra-long and highly accurate reads. Short-read Illumina data were used for genome survey, quality control, and polishing. Full-length RNA sequences obtained via Oxford Nanopore transcriptome sequencing support comprehensive gene model annotation. Additionally, Hi-C data provide chromosome-level scaffolding for constructing contiguous pseudomolecules. This integrated dataset supports a complete and accurate assembly of the C. sativa genome, enabling in-depth studies of genome structure, gene content, and functional genomics in this economically and medicinally important species.
