Vector capturing was used for experimental identification of potential off-target sites of 2 gRNA targeting alpha globin genes K562 cells were transduced with an Integrase-defective lentiviral vector expressing GFP and subsequently nucleofected with 30 pmol of Cas9:gRNA complex to induce double-strand breaks. Two weeks later GFP positive cells were sorted and integration sites were identified in this population using LM-PCR
