We present the de novo whole genome sequences of Streptomyces actuosus ISP-5337, Streptomyces sioyaensis B-5408, and Streptomyces cyaneus B-2296 sequenced by PacBio RSII circular consensus sequencing. Comparative genomics of the sequenced species with other Streptomyces in the NCBI refseq database shows the differences and similarities between these species. Bioinformatic analyses of encoded protein families coupled to secondary metabolite biosynthetic gene cluster predictions of each species identified multiple secondary metabolite biosynthesis-encoding clusters. We isolated secondary metabolite fractions from cultures, tested their antibiotic activity against Staphylococcus aureus ATCC 29213, and showed potent antibiotic activity from isolates of S. actuosus and S. sioyaensis. We characterized the fractions by liquid chromatography-mass spectrometry and showed the presence of thiopeptide metabolites. For future genetic engineering efforts, PacBio is the optimal method for sequencing of Streptomyces due to the depth of coverage and longer reads.
