Genome sequences of a novel Escherichia coli K-12 MG1655 L-form strain and its evolved variant

Escherichia coli K-12 MG1655 can grow as a cell wall-deficient L-form, which was subsequently subjected to long-term evolution. Replication of L-form bacteria without a cell wall requires specific genetic mutations.To investigate these mutations, we developed a new E. coli K-12 MG1655 L-form strain, evolved it over 800 generations in the absence of a cell wall, and conducted whole-genome sequencing.
