Single-cell profiling reveals Colonic CD8 Topography in IBD

Aims: Single-cell transcriptomic analysis of CD8 T cells in colonic tissue in Health and Ulcerative Colitis Results: We demonstrated significant heterogeneity within the CD8 population, defined disease associated populations and traced lineage relationships between different T cell subtypes Methods: Colonic biopsies from patients with endoscopically and histologically proven inflamed ulcerative colitis or healthy tissue were taken with informed consent. CD8 T cells were sorted using flow cytometry (FACS) and analysed using the 10X Single-cell RNA sequencing platform to yield matched gene expression, T-Cell receptor (TCR) and selected surface protein (CITE-seq) information across two batches (detailed below).
