An Agrobacterium isolate was extracted from the root of a mulaita plant (Sonchus cronutus) grown on the alluvial soil of Tutti Island in Khartoum State-Sudan. purified in five successive rounds of selection and identified at the National Institute for Promotion of Horticultural Export-University of Gezira, since 2005. It was subjected to physical, biological and molecular tests including colony characteristics, expression of enzymes, production of acids from carbohydrates, resistance to different herbicides and role of oxidase in such resistance, sensitivity to antibiotics, type of tumors as well as plasmid(s) characteristics. The herbicides pendimethalin, glyphosate and 2,4-D were used at dilution rates of 1:200, 1:1, 2:1 and 3:1 of herbicide: distilled water.. Cyanide was used as an oxidase inhibitor at dilution rates of 0.02, 0.2, 0.5 and 2.0 mg/l. It was used to inhibit oxidase activity in the bacterial suspension and on potato discs treated with the bacterium only and with herbicides. Next, this strain was used in potato disc bioassay to examine natural products for antitumor activity by determining the inhibition average percentage (IAP) of tumors for the different products. Latter, the whole chromosomal DNA of this bacterium strain was subjected to DNA sequencing by Illumina hiseq 1000/2500 technology.
