Gene expression profiles in the gut of DBA/1 mice whit spontaneus arthritis and treated with levofloxacion vs. those who were not received levofloxacin were compared. Levofloxacin induced the downexpression of inflammation-related genes and signaling pathways. Pools of total RNA were made per study group (six mouse per group): DBA1.SpA.Levo mice (test group) and DBA1.SpA.control mice (reference). The reverse transcription polymerase chain reaction (RT-PCR) was performed and the resulting complementary DNAs were labeled with Cy5 (DBA1.SpA.Levo) or Cy3 (DBA1.SpA.control). Hybridation analysis used the mouse 65-mer oligo library from Sigma-Genosys to compare the expression of 23,232 gene-specific oligonucleotide probes for mouse.
