Genomic DNA was extracted from the pancreas of the M4 fetus and the blood of its father and mother using a TIANamp Genomic DNA Kit (Tiangen, DP304). The genomic DNA (200 ng) was subjected to whole-genome sequencing at a sequencing depth of 30–40 × using the Illumina HiSeq 2000 platform at the Novogene Bioinformatics Institute (Beijing, China). Potential off-target sites (POTs) for each sgRNA were predicted to analyze site-specific edits according to Cas-OFFinder. Initially, all potential off-target sites in the mutant monkeys were compared with the reference genome. The filtered potential off-target sites were then compared to the parent genomes. After filtering, the final potential off-target sites were obtained and confirmed by Sanger sequencing
