Two varieties were used i.e. Anaerobic germination (AG) sensitive elite variety IR64 and AG-tolerant Ma-Zhan. Two treatments were assigned (1) seeds were either grown in air (control) on three layers of filter paper wetted with 1 ml of distilled autoclaved water in a 25 ml capped plastic tube (2) seeds were germinated in 6 cm depth (25 ml volume) of autoclaved distilled water in capped plastic tubes designated as AG treatment (hypoxia treatment). Tubes containing both control and AG treatment were incubatedat 30 C in the dark in an incubator for either 2 or 4d. Embryos were sampled at 2 days after sowing (DAS), and embryos and coleoptiles were sampled at 4 DAS. Embryos and coleoptiles were excised under a dissecting microscope Leica CLS 50X. For samples subjected to the AG treatment, embryos and coleoptiles were carefully dissected while submerged in water within a Petri dish. For the control samples grown in air, embryos and coleoptiles were excised on a single layer of wet filter paper. These dissected embryos and coleoptiles were snap frozen in liquid nitrogen and RNA was extracted these tissues.
