Gut microbiota plays a pivotal role in the development and function of the immune system, therefore it has been described as an important factor for the efficacy and safety of immunotherapy, including CAR-T. The mechanisms by which this occurs are not well understood. Short Chain Fatty Acids (SCFAs) are metabolites derived from the microbial fermentation that are involved in the regulation of metabolic, endocrine and immune function. In this study we analyze the transcriptomic changes that SCFA butyrate generates in CAR-T cells. Overall design: T cells were isolated from peripheral blood of four healthy donors and activated on anti-CD3/28-coated plates. After that, they were transduced with a CD19CAR lentiviral vector and separated into two conditions. One was exposed to SCFA butyrate and the other to the control medium. Comparative gene expression profiling analysis of RNA-seq data for both conditions was performed
