将胰腺癌手术标本中肿瘤部分分装成2-3mm大小的小块，经加入1%双抗的PBS溶液清洗5次后稍晾干水分放入培养皿中，加入特定的DMEM/F12培养基进行培养，培养条件为37℃，5%二氧化碳。培养24h后进行组织块换液，培养3-4天后镜下观察可见组织块周围有条状细胞爬出，继续培养至细胞布满整个培养皿底部时用0.05%胰酶消化5min，收集PSC细胞进行免疫荧光鉴定。
