Single cell RNA-Seq analysis of irradiated and non-irradiated skin cells in a nuclear accident patient.

This study aims to compare mRNA expression between irradiated and non-irradiated people’s skin by single cell RNA-Seq.The patient was a nuclear accident patient exposed to an X-ray spectrometer. In this study, the skin tissues was isolated from patient's different parts. The Control group of skin was collected from the patient belly without radiation for RNA-seq. The skin of IR group was collected from the irradiated skin tissue on the right hand of patients during skin grafting. Single cell RNA-Seq analysis of skin tissue from irradiated and non-irradiated parts of nuclear accident patient. ScRNA sequencing libraries were further generated by the 10x Chromium Single Cell 3′ Library & Single Cell 3’ v3 Gel Beads (10x Genomics). Briefly, gel beads with cell barcodes and skin cell suspension were wrapped in liquid droplets by a microfluidic platform (10x Genomics). A droplet containing single cells was collected and then lysed to generate Gel Bead-In Emulsions (GEMs).
