This dataset contains the raw confocal and western blot images for the publication "A mycobacterial Sec61 inhibitor disrupts lysosome function by blocking Vacuolar-ATPase biosynthesis", Hall et al. European Journal of Cell Biology, 2026 which describes the impact of the bacterial toxin mycolactone on lysosomal function. Mycolactone inhibits the Sec61 translocon, causing mislocalisation of membrane and secretory proteins to the cytosol. Cells respond by upregulating the proteasome and autophagy pathways but the down regulation of the V-ATPase accessory proteins ATP6AP1 and ATP6AP2 leads to progressive loss of acidic lysosomes, limiting the ability of cells to restore homeostasis.
