Single-cell transcriptomic analysis (scRNA-seq) of healthy adult human parietal pleura

The pleural lining of the thorax regulates local immunity, inflammation and repair. A variety of conditions, both benign and malignant including pleural mesothelioma, can affect this tissue. A lack of knowledge concerning the mesothelial and stromal cells comprising the pleura has hampered the development of targeted therapies. Here we present a comprehensive single cell transcriptomic atlas of the human parietal pleura. Healthy adult human parietal pleura were obtained from patients undergoing video-assisted thoracoscopic surgery (VATS) for pneumothorax. Tissue was minced, digested, filtered, and red blood cells were lysed. Dead cells and cellular debris were removed by Fluorescence-activated cell sorting (FACS) and live cells were submitted for scRNA-seq.
