We investigated implications of histone H3K79 methylation in cortical and development and dysfunction. DOT1L mediates H3K79me2/3, but little is known about the precise biological function of DOT1L in mammals. In order to understand the role of H3K79me2 and DOT1L during CNS development, we analysed primary neural cells derived from E14.5 NMRI. As it is unclear whether H3K79me2 is associated with an activating or repressing function on gene transcription, we revealed the genome wide pattern of H3K79me2 via ChIP-seq. We will correlate this data to the transcriptome to assess for the gene-specific influence of DOT1L in transcriptional regulation.
