To understand DIC-induced myeloid skewing of HSPCs, we profiled the transcriptome of LSK from DIC and control mice. Overall design: Mice were divided into 2 groups (3 per group): DSS-induced colitis (DIC) and control. DIC mice received 2.5% DSS for 5 days. For bone marrow (BM) analysis, femurs were flushed with ice-cold PBSF and filtered to obtain a single-cell suspension. LSK (Lin- Sca1+ cKit+) cells from BM were sorted by FACS Aria II and resuspended in PBS with 0.04% BSA.
