The exosomes secreted by SHED can be endocytosed by BMSC, and can partially reverse the inhibition of glucocorticoids on the viability and osteogenic differentiation of BMSC. Transcriptome data analysis combined with qPCR showed that three pathways, Rap1, PI3K-Akt and CAMs (Cell adhesion molecules), may be related to this experimental phenomenon. Primary dental pulp cells were isolated and cultured from human deciduous tooth pulp, and SHED was isolated and purified by limiting dilution method and exosomes were isolated from SHED cell supernatants by ultracentrifugation. Cell surface markers CD31, CD34, CD45, CD73, CD90 and CD105 were detected by flow cytometry. ALP and Alizarin Red staining were used to identify osteogenic differentiation ability, and exosomes were identified using TEM (transmission electron microscopy), NanoFCM and Weatern Blot. PKH67 fluorescent labeling tracked the uptake of exosomes by BMSC. Transcriptome combined with qPCR to explore its mechanism.
