RNA seq and functional annotation of Sarcoptes scabiei var. nyctereutis

Sarcoptes scabiei is a widespread, highly contagious skin disease that affects many mammals. However diagnosis is difficult and there is limited data available regarding the molecular basis of host specificity and host-parasite interactions. Therefore, we performed transcriptome analysis of Sarcoptes scabiei for basic research and clinical diagnostic. In this study, total RNA was extracted from livie mites (Sarcoptic scabiei var. nyctereutis) from crusted Japanese raccoon dogs (Nyctereutes procyonoides) for quality assessment, RNA-Seq, functional annotation, and coding region (CD) prediction and verification. The Illumina platform generated 320.80G clean reads and 110,911 genes with 734.48T were assembled. In total, 110,346 genes were annotated in databases. Finally, 44,773 CDs were detected in the 110,033 genes, of which 27,076 complete CDs were matched with Sarcoptes manually in non-redundant nucleotide. This study successfully completed the RNA-Seq and functional annotation of Sarcoptic scabiei var. nyctereutis for the first time, which provides molecular data for future studies on the identification and pathogenic genes of Sarcoptidae.
