This study seeks to test how PILRA KO affects human microglial response to AD pathology in vivo using a xenotransplant mouse model of AD and single-cell RNA-sequencing. Overall design: WT and PILRA KO iPSC-derived HSCs were transplanted into immune-deficient mice in a WT or 5XFAD AD model background and allowed to mature into microglia for 6 months. Cells were harvested and FACS sorted then single-cell libraries were built and sequenced.
