Recent evidence has indicated the significant role of exosomes in osteoporosis (OP) both in vivo and in vitro, and the plant-derived exosome-like nanovesicles (ELNs) have gained huge attention as potential therapeutic effects. In current study, the isolation and purification of ELNs from Panax notoginseng (sanqi) was performed successfully, and the internalization of ELNs by human bone marrow mesenchymal stem cells (BMSCs) positively affect their viability. Moreover, the ELNs can inhibit osteoclast activity in vitro and inhibit OP in an ovariectomized mice model, suggesting that sanqi-derived ELNs could be used to reverse the OP. Subsequently, the experiments including miRNA sequencing, RNA sequencing and dual luciferase reporter gene assay, indicated that ELNs enriched many miRNAs, which might regulate the differentiation of BMSCs. Furthermore, it was proved that ELNs could effectively load the miRNA-155 and reverse the OP more effectively than normal ELNs. Collectively, our data suggested that sanqi-ELNs might be utilized as not only a natural medicine and a non-toxic nanocarrier of drug for the OP treatment.
