Primary fixation included: 2.5 % glutaraldehyde, 2% formaldehyde in 0.1 M Na-phosphate buffer, pH 7.4. Post-fixed in 2% OSO4 in 0.1 M Na-phosphate buffer, pH 7.4. Stained en bloc in 1% uranyl acetate. The tissue was then dehydrated in a graded series of ethanol and infiltrated with Spurr's resin. Thin sections of 70 nm were trimmed using a diamond knife and post-stained in uranyl acetate and lead citrate. This micrograph image was taken using a Phillips CM 100 transmission electron microscope at an accelerating voltage of 80kV.前固定液配方为：0.1mol/L磷酸钠缓冲液（pH 7.4）中含2.5%戊二醛、2%甲醛。随后使用0.1mol/L磷酸钠缓冲液（pH 7.4）配制的2%四氧化锇（OSO4）进行后固定。接着采用1%乙酸铀酰对组织进行整体块染。之后将组织经梯度乙醇系列脱水，并以Spurr树脂完成渗透处理。使用钻石刀修切70nm超薄切片后，以乙酸铀酰和柠檬酸铅进行后染色。本显微图像采用飞利浦CM 100型透射电子显微镜拍摄，加速电压为80kV。
