Thyroid hormone (3,5,3'-triiodothyronine, T3) sensitively influences the pituitary gland, a source of hormones that control tissues throughout the body. The underlying transcriptional response is believed to hinge crucially on interaction of T3 receptors with enhancers in the genome but it remains unknown how T3 regulates pituitary chromatin and how this regulation adjusts to hypothyroid and hyperthyroid conditions. Overall design: The binding sites of the biotinlyated thyroid hormone receptor beta (Thrb) at mouse tissue under either euthyroid, hypothyroid or hyperthyroid conditions by ChAP-seq. ChIP-Seq profiling of histone H3K27ac,H3K4me1 and H3K4me3 by ChIP-Seq and ChAP-seq profiling of Thrb by ChAP-seq at mouse pituitary or brain under either hypothyroid or hyperthyroid conditions. The chromatin accessibility profiling by ATAC-Seq at Thrb2 positive mouse pituitary cells under either hypothyroid or hyperthyroid conditions. The gene expression profiling by RNA-Seq at mouse pituitary under either hypothyroid or hyperthyroid conditions.
