Peroxisome elevation induces stem cell differentiation and intestinal epithelial repair

Epithelial-repair-dependent mucosal healing (MH) is associated with a morefavorable prognosis for patients with inflammatory bowel disease (IBD), and lowersrelapse, hospitalization, and surgery. However, the ill-defined mechanism underlyingMH has made it difficult to develop therapeutics that induce epithelial repair and MHin IBD patients. We found a striking upregulation of a highly plastic organelle, theperoxisome, in the injured crypts of IBD patients. By leveraging the RNA sequencing on Drosophila, we demonstrated that peroxisome elevation enhanced RAB7-dependent late endosome maturation, which in turn promoted the stem/progenitor-cell differentiation via modulation of JAK/STAT-SOX21A signaling. This, in turn, enhanced ISC-mediatedregeneration. Here, this project stored the raw sequencing data associated with this study.
