Extensive studies in plants have revealed how small interfering RNA (siRNA) mediated DNA methylation functions in epigenetic transcriptional repression. Briefly, two types of siRNA 21 22 nucleotide (nt) or 24 nt in length are understood as the major siRNAs that direct DNA methylation. Specifically in plants, siRNAs are produced from transposons and transgenes. To identify the DNA methylation status, we performed bisulfite sequencing of heterozygotes, initial homozygotes, and successive homozygotes of the first trifoliolate sampled at V2 stage.
