The deposited data are from experiments that support a manuscript submitted by Sherley et al., 2021-2022. The purpose of the manuscript was to continue validation of kinetic stem cell (KSC) counting, a first method for routine determination of the stem cell-specific fraction and dosage of tissue stem cell-containing cell preparations. The tissue stem cell focus for this dataset is human hematopoietic stem cells (HSCs). KSC counting is a computer simulation technology. The input for KSC counting software is cumulative population doubling (CPD) data and dead cell fraction data from replicate serial cultures. The data set includes the input total cell count data, dead cell fraction data, and CPD data from replicate (2 or 3) serial cultures of different commercial suppliers of three different human tissue cell donor sources containing HSCs. These include CD34+-selected umbilical cord blood, mobilized peripheral blood cells, and CD34+-selected mobilized peripheral blood cells. The KSC counting analyses reported based on these data were also used to validate rapid counting Rabbit algorithms. Rabbit algorithms allow calculation of stem cell-specific fraction or dosage from CPD data or population doubling time data without serial culture.
