Single-cell gene expression of neocortex after transient Yamanaka factor expression during brain development

Yamanaka factors (YFs) can reverse some aging features in mammalian tissues, but their effects on the brain remain largely unexplored. Here, we used single cell RNA sequecing (scRNA-seq) to analyze the effects of YFs induction during cortical development. We used the conditional inducible-four factors (i4F) mouse line, which allows the induction of YFs in a Tet-ON system. This line was crossed with a Cre-dependent floxed rtTA and the nervous system-specific Nestin-Cre (Nes) mouse line. In this mouse model (i4F-Nes), YF expression was induced by administering doxycycline from E10.5 to E14.5. The rostral region of the mouse neocortex was collected at E15.5. Two i4F-Nes samples and two littermate control (CTR) samples were prepared for scRNA-seq analysis.
