Characterization of isolated bacterial genomic DNA in microfluidic chambers (Genome-in-a-Box) - Dataset

Data underlying the research presented in my thesis, including a manuscript currently in preparation "A microfluidic platform for extraction and analysis of bacterial genomic DNA". Including fluorescence and Mass Spec.<br>The data contains raw and processed 3D time resolved fluorescent images of spheroplasts and nucleoids in microfluidics traps. The data has been split at each time point to individual planes, such that all tiff files are 2D. The dataset includes all object detection, segmentation, and quantification results (i.e. all intermate steps of analysis). The data allowed for tracking spheroplasts trapping on the device, their lysis, nucleoid expansion, and analyzing various properties such nucleoid size, structure and dynamics at range of conditions (e.g. addition of crowding molecule or a DNA binding protein). The data can be loaded and analyzed with tools provided at this Zenodo repository.

本论文所呈现研究的配套数据集，包含一篇正在撰写的手稿《用于细菌基因组DNA提取与分析的微流控平台》（A microfluidic platform for extraction and analysis of bacterial genomic DNA），以及荧光数据与质谱（Mass Spec）数据。该数据集涵盖微流控陷阱中原生质球（spheroplasts）与类核（nucleoids）的原始及经处理的三维时间分辨荧光图像。所有数据已按每个时间点拆分为单独成像平面，因此所有TIFF文件均为二维格式。本数据集包含全部目标检测、图像分割及定量分析结果，即分析流程中的所有中间步骤。依托该数据集，可实现对装置内捕获的原生质球、其裂解过程与类核扩张行为的追踪，并可在多种实验条件（如添加拥挤分子或DNA结合蛋白）下，分析类核的大小、结构与动态变化等多项特性。用户可通过本Zenodo仓库提供的工具加载并分析该数据集。
