The immune response differences of rats induced using RBD3-Fc mRNA vaccines inoculated intradermally by NFIS and intramuscularly by needles

We have employed a single cell sequencing approach using 10× chromium microfluidic system to further illustrate the effect of inoculation routes on T cell immune responses induced by the mRNA-LNP vaccine and naked mRNA vaccine. Rat splenocytes were harvested at week 6. CD45+ splenocytes were separated and captured as immune cells using flow cytometry, then analyzed using scRNA-seq. THis study revealed the advantages of NIFS delivery, which evoked potent T cell responses.本研究采用基于10× Chromium微流控系统的单细胞测序方案，以进一步探究接种途径对mRNA脂质纳米颗粒（mRNA-LNP）疫苗与裸mRNA疫苗诱导的T细胞免疫应答的影响。研究人员于第6周收获大鼠脾脏淋巴细胞，通过流式细胞术分离并捕获作为免疫细胞的CD45阳性脾脏淋巴细胞，随后采用单细胞RNA测序（single-cell RNA sequencing, scRNA-seq）进行分析。本研究揭示了NIFS递送方式的优势，其可诱发强效的T细胞免疫应答。
