Genome-wide DNA methylation profiling was performed to identify novel markers for DNA methylation-based identification of forensically forensically relevant body flulids. CpGs specifically methylated or unmethylated in saliva, vaginal swabs, blood and semen were searched by comparing beta values of about 850000 CpGs of pooled samples of each body fluid. Two sets of pooled samples of extracted DNA from each body fluid type (saliva, blood, semen, or vaginal swab) were prepared for genome-wide methylation analysis by Illumina Infinium MethylationEPIC. Each pooled DNA sample consisted of equal amounts of DNA from three individuals. Five hundred nanograms of each pooled sample were bisulfite-converted and analyzed according to the manual protocol of Illumina Infinium HD Methylation assay.
