To identify the most important gene(s) for maintaining the stem cell properties of leukemia stem cells (LSCs), we analyzed the transcriptional profiles of LSC and non-LSC subsets from acute leukemia patients and identified S1PR1 as a novel biomarker of LSCs. Then to understand the underlying mechanisms of S1PR1-dependent stemness maintenance, we performed gene expression profiling analyses comparing CD34+S1PR1+ and CD34+S1PR1- cell fractions from AML patients and healthy donors; we also determined the global transcriptional response of KG1 cells to an inhibitor of S1PR1 (FTY720) at two concentrations. Data set 1: Three acute myeloid leukemia (AML) samples (patient #M1, #M2, and #M3) were sorted into paired LSC (CD34+CD38-) and non-LSC (CD34+CD38+, CD34-CD38+, and CD34-CD38-) fractions by flow cytometry. RNA-seq profiling was performed on the paired fractions for each individual. Data set 2: Three Philadelphia chromosome-positive B cell acute lymphoblastic leukemia (ALL) samples (patient #L1, #L2, and #L3) were sorted into paired LSC (CD34+CD38-CD58-) and non-LSC (CD34+CD38+CD58+, CD34+CD38+CD58-, and CD34+CD38-CD58+) fractions by flow cytometry. RNA-seq profiling was performed on the paired fractions for each individual. Data set 3: RNA-seq profiling was performed on CD34+S1PR1+ (labeled as P_pos) and CD34+S1PR1- (labeled as P_neg) bone marrow mononuclear cell fractions from AML patients (patient #9 and #10) purified by flow cytometry. Data set 4: RNA-seq profiling was performed on CD34+S1PR1+ (labeled as D_pos) and CD34+S1PR1- (labeled as D_neg) bone marrow mononuclear cell fractions from healthy donors (donor #1 and #2) purified by flow cytometry. Data set 5: RNA-seq profiling was performed on KG1 cells that were pre-treated with FTY720 at 3 μM (labeled as F3) or 4 μM (labeled as F4) for 24 h or sham treatment (labeled as F0) without FTY720.
