Our study includes a high-throughput sequencing approach of soil and stone samples from Namibian drylands. We paired culture-independent 16S rRNA gene amplicon sequencing with culture-dependent (isolation of bacteria) technique to assess community structure and diversity patterns. We modified the DNA extraction method for stone samples and separated into intra- and extracellular DNA.
