we prepare pre-activated macrophage-like nanoparticles that carry specific receptors for Mycobacterium tuberculosis (M. tb), which allows them to target tuberculous granulomas and internal M. tb, simultaneously. The reason we selected M. m instead of M. tb to stimulate macrophages in this study is that M. m can be handled in a biosafety level II laboratory, which facilitates massive production of Nanoparticles. More importantly, M. m is the closest genetic relative of M. tb and both species contain nearly the same antigens and virulence determinants, and thus can activate macrophages to overexpress identical pathogen-associated receptors for specific targeting of M. tb. Gene expression related to M. tb on pre-activated RAW264.7 macrophages were analyzed through transcriptome sequencing.
