Using photoactivatable ribonucleoside crosslinking and biotinylated Cp retrieval, multiple binding sites on viral gRNA where identified. Vero-BirA cells were infected with SFV-Cp-mAVI for 1.5 h and treated with biotin and photoactivatable ribonucleoside 4-thiouridine (4SU). Cp-mAVI-biotin was retrieved form Total cellular lysates, purified Cellular Nucleocapsids (NC) and from purified Viral NC with Streptavidin beads, and crosslinked RNAs where further processed using the standard PAR-CLIP protocol
