The resting cyst of dinoflagellates is usually the zygotic stage of their life cycles and results from sexual reproduction. Resting cysts in sediments can resist harsh environmental conditions and are protected from viruses, grazers or parasite attacks. As a crucial part of the ecology of HAB-forming dinoflagellates, resting cyst has been proven to be important in genetic recombination, bloom initiation, bloom termination, bloom recurrence, and geographic expansion. However, gap remains in our knowledge about physiological and molecular mechanism underlying the resting cyst formation in dinoflagellates. The thecate dinoflagellate Scrippsiella trochoidea is a cosmopolitan alga that commonly forms blooms in many regions around the world. In this study, high-throughput paired-end RNA-sequencing (RNA-Seq) technology was applied to transcriptomes of Scrippsiella trochoidea vegetative cell and resting cyst samples, respectively. Comparative analysis of gene expression was designed to correlate the effect of physiological mechanisms on the molecular level.
