Role of TBP in reactivation of transcription following mitosis [RNA-Seq]

We performed nascent chromatin-associated RNA analysis as a time course following mitosis to determine how transcription is reactivated globally as cells enter G1. Overall design: We used asynchronous mouse embryonic stem (ES) cells, or Nocodazole-synchronized mitotic ES cells as starting material. Mitotic cells were then released into fresh media for 30 or 60 minutes to allow entry into G1. After extracting nascent chromatin-associated RNA, we used stranded single end sequencing and mapped the reads to the mm10 genome.
