The goal of this study was to investigate the DNA methylation and gene expression changes associated with melatonin deficiency in plants and their impact on leaf senescence and grain yield in rice. Melatonin is a phytohormone known to play important roles in plant growth and stress responses, but its specific effects on DNA methylation and leaf senescence were not well understood. In this study, we discovered that melatonin deficiency in rice plants resulted in premature leaf senescence during the grain-filling stage, leading to a decrease in grain yield. Whole-genome bisulfite sequencing and RNA-seq techniques were used to uncover the potential mechanisms underlying this premature leaf senescence. The results of this study provided insights into the epigenetic mechanisms underlying melatonin-regulated leaf senescence.
