In aquaculture, intestinal inflammatory disease induced by replacing fish meal with high level of soy protein is a common phenomenon. Pearl gentian grouper, a kind of marine fish with important economical and nutritional value, exhibited the similar problem. To the best of our knowledge, few full-length grouper transcriptomes are available. In this study, a library composed of 7 different treated hindgut tissues including fish meal (FM) control group, 20% soybean meal substituted for fish meal (SBM20), SBM40, 20% soybean protein concentrate (SPC20), SPC40, 20% fermented soybean meal substituted for fish meal (FSBM20) and FSBM40 was constructed and sequenced using Pacific Biosciences single-molecule real-time (SMRT) long-read sequencing technology. A total of 420006 full-length non-chimeric reads were obtained, from which 82351 high-quality transcripts were obtained after error correction and sequence clustering and redundant. In addition, a total of 77815 transcripts were annotated against protein database (Nr, Nt, Pfam, KOG, GO, Swiss-Prot and KEGG database). A total of 49093 long non-coding RNAs (LncRNAs) and 141702 simple sequence repeats (SSRs) were identified in this study. Meanwhile, the isoforms of many genes were also identified in this study. This study provides new and valuable reference transcripts, which will greatly facilitate further exploration of pearl gentian grouper.
