Chronic kidney disease (CKD) affects more than 10% of the global population with limited effective therapies. Understanding the underlying mechanisms of kidney fibrosis, the common final pathway of CKD to kidney failure, is essential to the development of new therapeutic strategies. Activation of the complement system and excessive production of the anaphylatoxin C5a observed in CKD suggests its possible role in disease pathogenesis and progression by promoting inflammation-driven kidney injury. This study aims to investigate the diverse roles of C5a receptor 1 (C5aR1) in two models of progressive CKD and provide evidence on C5aR1 antagonism as a therapeutic strategy.
