Eastern water dragon archival sequencing - second collection

The data consist of 150 bp paired-end Illumina sequencing reads resulting from hot alkaline lysis extraction of 14 archival eastern water dragon (Intellagama lesueurii lesueurii) specimens. The specimens, collected between 1925 and 2008, were spirit-preserved and all but two were formalin-exposed. Liver or muscle tissue was dissected and subjected to hot alkaline lysis as previously described (Campos & Gilbert, 2012) with minor protocol adjustments. The extracted genomic DNA was then prepared for sequencing using an IDT xGEN Prism library prep kit. These data accompany a successful pilot run testing the efficacy of the IDT xGEN prism in preparing archival DNA for sequencing. These data further test the performance of hot alkaline lysis in recovering genomic DNA from specimens across a range of preservation conditions. The data were generated as part of a collaboration with Celine Frere at the University of the Sunshine Coast, as part of a reciprocal agreement for early access to the reference genome sequence of Intellagama lesueurii.
