This study shows that a global view of DNA methylation-deregulated lncRNAs in 12 TCGA cancer types. We further demonstrate that a hypomethylated promoter-mediated activation lncRNA, DMDRMR, as a cofactor for IGF2BP3 to stabilize their target genes in an m6A-dependent manner, i.e. CDK4, thus exerting essential oncogenic roles in ccRCC. These findings provide lncRNA as m6A-reading cofactor in cancer and suggest DMDRMR/IGF2BP3/CDK4 axis is a key signaling pathway for ccRCC.
