A total of 9 clinically healthy Simmental bulls were selected and used to conduct the Direct RNA sequencing. Nanopore ONT sequencing can be used to characterize the transcriptome complexity of testis in Simmental bulls at three developmental stages (Prepuberty, Puberty, and Postpuberty). We conclude that DRS enables the identification of complexity and dynamics of the bovine transcriptome. The comprehensive annotation of transcript isoforms further helps to refine the bovine genomics database and identify the candidate genes or transcripts related to testicular development and spermatogenesis.
