Recent research has shed light on the neuroimmunomodulation of macrophages, but the precise underlying mechanisms remain poorly understood.The maintenance of homeostasis in various organizations requires neuroimmune communication between the sympathetic nervous system and the immune system. The sympathetic nervous system adjusts macrophage phenotype via the adrenergic pathway.This study aimed to delineate the molecular mechanisms linking adrenergic pathway to RAW264.7. Overall design: To examine the impact of adrenergic pathway on the regulation of RAW264.7, we categorized the RAW264.7 cells into three groups. The first group was stimulated solely with IL-4 to induce M2 polarization. The second group was stimulated with IL-4 along with the ß2 adrenergic receptor agonist isoproterenol (ISO), activating the ß2 adrenergic signaling pathway in RAW264.7 cells. The third group was pre-treated with the ß2 adrenergic receptor antagonist ICI118,551, followed by stimulation with IL-4 and ISO, effectively blocking the ß2 adrenergic signaling pathway in RAW264.7 cells. This approach aimed to elucidate how adrenergic pathway influences the dynamics of RAW264.7.
