In plants, the miRNA biogenesis pathway is regulated by the cooperation of several host factors. Being one of a core components of the miRNA processer, it is konwn that the Arabidopsis thaliana HYPONASTIC LEAVES1 (HYL1) preferentially binds and helps to process the primary miRNA transcripts from long double-stranded RNA (dsRNA) into small mature miRNAs. Besides, HYL1 also participates in plant growth, drought resistance, light response, hormone signaling pathway, fungal and bacterial resistance. Rice double-stranded RNA binding protein (dsRBP), DRB1, is the homologous to HYL1. However, few studies about DRB1 have been investigated and its function in rice is still unknown. Additionally, as a dsRBP, the detailed binding map of DRB1 remains a mystery. To address these issues, we applied TRIBE (Targets of RNA-binding proteins Identified By Editing) method to rice DRB1. Leave and root samples were used to explore a organism-specific binding map of DRB1. The substrate RNA with the A-to-G RNA editing events were further identified by RNA sequencing and a dsRNAome targeted by DRB1 was obtained. The functions of DRB1 in rice were further investigated.
