This assembly represents a combination of transcripts from different developmental stages of Ascaris. Thus, six libraries from embryo, larvae, testis, ovary, complete female reproductive system, and whole males were prepared. Reads from each of the 6 cDNA libraries were initially assembled with Velvet (Zerbino and Birney 2008). A set of reference sequences were built by combining all these assemblies together (phrap), and each library was reassembled (with Velvet) to obtain an improved assembly. Thus, an improved reference sequence was built, and this cycle was iterated until no further improvement could be achieved. The final assembly was improved by scaffolding and by experimental verification on genes of interest.
